Chromosome engineering allows deletion or duplication of large part of a chromosome.
This technology is specially designed to generate monosomic and trisomic syndromes in the mouse.

Cre/loxP-mediated chromosome engineering of the mouse genome.

Brault V, Besson V, Magnol L, Duchon A, Hérault Y.

Handb Exp Pharmacol. 2007;(178):29-48.

ICS expertise:

  • For large genomic region two targeting events are required that could be used to generate using the Cre recombinase, cis or trans mediated recombination leading to deletion or duplication of the region of interest.
  • Inversion or translocation could be induced.

Risks:

  • Decrease efficiency due to increased distance between loxP sites.
  • Potential haploinsufficiency of the deletion may be compensated by the generation of tandem duplication for the region of interest.
